چکیده

The structure of carbohydrate chains of dog gastric mucus glycoprotein bearing Forssman antigenic determinant was investigated. The reduced oligosaccharides obtained by alkaline borohydride reductive cleavage of the glycoprotein were chromatographed on a immunoaffinity column containing monoclonal anti-Forssman antibody coupled to cyanogen bromide-activated Sepharose 4B. The bound oligosaccharides, eluted from the affinity column with 6 M urea, separated on thin layer chromatography into two major components. These two compounds, representing 92% of the absorbed oligosaccharides, were purified to homogeneity, and their structures and antigenic properties were determined. Based on the results of immunological assays, analysis of sugar composition, sequential degradation with specific glycosidases, and methylation analyses of the intact and enzyme-degraded compounds, we propose the following structures for these oligosaccharides. (formula, see text).

A Novel mAb against a Human CD34 Peptide Reacts with the Native Protein on CD34+ Cells

Background: Human CD34 is a transmembrane glycoprotein which is expressed in human hematopoietic stem cells (HSCs) and the small- vessel endothelial cells of a variety of tissues. CD34 plays a critical role as a marker for diagnosis and classification of leukemia. Anti CD34 antibodies are used for isolation and purification of HSCs from bone marrow, peripheral blood and cord blood. Objective: ...

Peripheral alpha-linked N-acetylglucosamine on the carbohydrate moiety of mucin derived from mammalian gastric gland mucous cells: epitope recognized by a newly characterized monoclonal antibody.

To obtain a tool to study the structural characterization and the detection of mucin derived from the gastric gland mucous cells, we developed a monoclonal antibody, designated HIK1083, against mucin purified from rat gastric mucosa. In an ELISA, HIK1083 reacted strongly with the mucin purified from a deep layer of the corpus and antrum but only slightly reacted with that obtained from the surf...

Production, Purification and Characterization of Chicken Egg Yolk Monoclonal Antibody Against Colonization factor antigen -1 of Enterotoxigenic Escherichia coli Causing Diarrhea

Enterotoxigenic Escherichia coli (ETEC) causes diarrhea in both humans and animals. The contaminated food and water are the most common vehicles for ETEC infection. The colonization factor antigen (CFA-1) is a fimbriae protein that promotes adherence of the ETEC strain to the epithelium of the small intestine of the host. In this study IgY proteins were produced against the CFA-1 of ETEC in imm...

PRODUCTION AND CHARACTERIZATION OF MONOCLONAL ANTIBODIES TO CLOSTRIDIUM PERFRINGENS ENTEROTOXIN DERIVED FROM DIFFERENT ANIMAL SPECIES

The enterotoxins (ETs) of Clostridium perfringens isolated from enterotoxemic or diarrheic alpacas, pigs, calves, dogs, and horses were obtained from sporulated cell extracts. The ETs from alpaca, pig, and calf isolates were chromatographed on Sephadex G-lOO. Monoclonal antibodies (MAbs) against ETs derived from alpaca, pig, and calf C. perfringens isolates were produced. The MAbs were used...
